Purificação parcial e caracterização de uma lectina de sementes de Crotalaria retusa L.

Lectinas são proteínas ou glicoproteínas de origem não imune, que se ligam reversivelmente a carboidratos específicos e são amplamente distribuídas na natureza. Esse trabalho teve como objetivo a purificação parcial e caracterização de uma lectina de sementes de Crotalaria retusa L. (CrL). O extrato bruto foi obtido através da trituração das sementes e posterior homogeneização em solução de NaCl 0,15M. A lectina foi capaz de aglutinar todos os eritrócitos humanos testados, com uma concentração mínima de 7,81μg/ml. CrL foi inibida por L-fucose, N-acetilgalactosamina e D-galactose nas concentrações mínimas de 12,5mM, 25mM e 25mM, respectivamente, mas não foi dependente de cátions divalentes. CrL foi parcialmente purificada através de cromatografia de exclusão molecular em coluna de Sephadex G-75 e exibiu uma massa molecular de 30 kDa em eletroforese em gel de poliacrilamida (SDS-PAGE). A lectina foi estável até 100oC por 30 minutos e a atividade hemaglutinante foi mantida entre o pH 7 e 9.
