Statistical analysis applied to comparison of methods for determining the pKa of acid-base indicator by UV-Vis spectroscopy

The pKa defines the predominant protolytic species present in solution as a function of the pH. This impacts several physicochemical properties, especially the visual color changes of the solution for acid base indicators. Such changes can be microscopically monitored by ultraviolet/visible spectroscopy. This paper presents a statistical comparison of graphical and analytical approaches of pKa determining for acid-base indicator bromocresol green (VB) through its absorption spectra as a function of pH. The methodology consisted of chemometric quantifying of the coexisting species in solution and determining pKa by three different methods: (I) intersection of linear curves, (II) intersection of axis and (III) analytical. The results showed that two variables explain 99% of the variance contained in the VB data, indicating the coexistence of two species in equilibrium and a single transition as a function of the pH. The pKa values were 4.22±0.05; 4.75±0.12 and 4.50±0.02 by methods I, II and III, respectively. Statistical analysis indicated that method I was significantly different from the others, method II was the most accurate and method III is the most precise. It should be noted that statistics put along with experimentation can improve the interpretation of analyzes and scientific knowledge across-the-board.
