Síntese e avaliação in silico do potencial farmacológico de 1,2,4-oxadiazol 3,5-dissubstituído

Resumo

Os heterociclos oxadiazólicos estão presente em inúmeras estruturas com diversas propriedades  farmacológicas. Contudo, o anel heterocíclico de 1,2,4-oxadiazol de cinco membros tem recebido atenção  considerável por causa de suas propriedades bioisostéricas únicas e um espectro excepcionalmente amplo de atividades biológicas. A literatura reporta um número crescente de publicações envolvendo 1,2,4- oxadiazol ao longo dos últimos anos, as quais tem tido como foco seu diversificado potencial  farmacológico. Neste trabalho, é descrito a síntese, caracterização estrutural e avaliação in silico de 1,2,4- oxadiazois. As (Z)-4-fluormetil benzamidoxima (3a) e a (Z)-3-(trifluormetoxi)-benzamidoxima (3b),  produtos de partida de interesse, foram obtidos em bons rendimentos (85% 3a e 93% 3b). Por outro lado,  os 2-(3-aril-1,2,4-oxadiazol-5-il)-3,4-(metilenodioxi)-cinamila (6a-b) foram obtidos a partir do trans-3,4- (metilenodioxi)-cinamato de metila e arilamidoxima através da irradiação em forno de micro-ondas  focado rendimentos de 88 e 65%, respectivamente. Além disso, o estudo in silico indicou que todos os  compostos sintetizados apresentam baixo risco de toxicidade crônica e podem ser administrados por via  oral. Além disso, existem fortes indícios físico-químicos que sustentam a probabilidade desses compostos apresentarem atividade ansiolítica, antidiabética, antiemética e antiprotozoária.
